Figure 2.  Avl9 was identified in a triple-synthetic lethal screen for mutants that are lethal in combination with the vps1 apl2 mutations.  In an analogous chemical genetic screen, we are identifying small molecules that inhibit the growth of the double-mutant but not to the corresponding wild-type strain.  Some of our compounds have much less effect on single mutants (A) whereas others inhibit the growth of one or both single mutants in addition to the double mutant.  Many of our compounds block secretion.  At least one such compound results in rapid (within 30 min) accumulation of abnormal membranes in yeast cells, as shown in this thin-section electron micrograph (B).
